This project was designed to find out cGMP-dependent ion channel required for the photoresponse in planaria by inhibiting the expression of a gene, the gene for the cGMP-gated ion channel, which is known to be required for phototransduction in numerous organisms, the gene will be cloned and used in the RNA interference process. Identification of homologues of the cGMP-gated ion channel in Schmidtea mediterranea. In this research we isolate of total RNA from planarian and synthesized the cDNA. The amplified DNA will be cloned into a standard vector by using p-GEM-T easy vector kit. The gene will be transferred to a vector that allows for the synthesis of dsRNA. The dsRNA will be added to liver paste (a food source) and fed to the worms. The worms will be tested for the ability to respond to bright light.
